The Ettamogah Pub near Albury is for sale with price expectations of more than $3 million.
The Ettamogah Pub was originally part of a cartoon created by Ken Maynard in Australasian Post magazine.
In 1987, businessman Lindsay Cooper built an actual pub in Ettamogah, which opened in 1987. It features a distinctive architectural style true to the original cartoon design.
Further Ettamogah Pubs were later built in Sydney in NSW, Palmview on the Sunshine Coast in Queensland, and Cunderdin in Western Australia.
Brokerage firm Manenti Quinlan and Associates is handling the sale of the original Ettamogah Pub, with director Leonard Bongiovanni noting: "Rarely does an opportunity present itself to acquire an Australian icon along with the opportunity of trading under a distinguishable brand steeped in history."
Bongionvanni said the 4.81ha site offered "multiple areas to enhance revenue and become a village off the highway".
"Numerous value-add opportunities are available via the currently unused areas which require an insightful and hands-on operator to activate and take advantage off," he added.
"Opportunities of this calibre are rarely available to the open market."
In addition to the pub, the Ettamogah site has a "village" available with retail shops and a stand-alone bottle shop on the property.
"The Ettamogah site offers an astute operator the opportunity to create a village with five retail shops, a reception centre and large dining barn with structures close to completion and under-utilised which will provide further upside once fitted out," Mr Bongiovanni said.
The property will be sold by expressions of interest which will close on June 7.
